Recently, researchers at Stanford University School of Medicine have developed an antibody that targets CD47 and can safely and effectively treat five types of pediatric brain tumors. Cancer cells interact with SIRPα on macrophages by CD47, signaling a “don't eat meâ€. The anti-CD47 antibody, called Hu5F9-G4, blocks these interactions, prompting macrophages to phagocytose brain cancer cells without harming healthy brain cells. This achievement was published in the Science issue of Science Translational Medicine.
In fact, these anti-CD47 antibodies developed by Stanford researchers have been used in early clinical trials to test adult patients with tumors outside the central nervous system. However, until recently, they have not been tested for the efficacy of pediatric brain tumors.

In an in vitro cancer cell line and a xenograft (PDX) mouse model derived from a patient's tumor tissue, Hu5F9-G4 against five invasive pediatric brain tumors, group 3 medulloblastoma, SARS Atypical teratoid rhabdoid tumor, primary neuroectodermal tumor, pediatric glioblastoma, and diffuse true pons (diffuse intrinsic) Pontine glioma) - shows a significant inhibitory effect. In the trial of healthy mice receiving the latter high-grade glioma, the low-dose and high-dose Hu5F9-G4 extended the average survival of the mice from 21 days to 32 and 38 days, respectively.
“For many of these tumors, there is no cure at all,†said the author of the article, Professor Samuel Cheshier. “Defining a diagnosis is tantamount to a death sentence.â€
