Responsive hydrogel or treat arthritis

Recently, Nature-Communication published a mouse model and demonstrated a release gel that responds to the activity of inflammatory arthritis.

The disease activity of patients with inflammatory arthritis is low for a while, high for a while, and high when it is called a disease attack. Drug therapy with a constant delivery dose cannot cope with and adapt to these changes in activity, so optimal doses may not be achieved during the cycle of disease activity.

Jeffrey Karp of the Brigham and Women's Hospital in Boston, Massachusetts, and colleagues attempted to resolve a mismatch between disease activity and drug dose by developing a hydrogel that responds to seizures. In cells and mouse models of inflammatory arthritis, the gel decomposes when it comes into contact with arthritis-related enzymes.

The gel can be loaded with different small molecules that release the appropriate amount of drug depending on the severity of the onset of the disease. When the gel is loaded with a corticosteroid for the treatment of arthritis, a decrease in arthritis activity is observed.

Responsive hydrogels are a potential way to treat inflammatory arthritis, the researchers say. But they are also cautiously suggesting that more research is needed to optimize the composition of the gel and that it is necessary to test on large animals with human-sized joints before considering possible human clinical trials. The authors conclude that this may be the first step in designing a treatment plan that responds to disease activity.
