Northeastern University: Discovering a new mechanism for treating fatty liver

Release date: 2017-10-17

A few days ago, the students of Northeastern University discovered through experiments that the iron chelator DFI has a significant alleviating effect on nonalcoholic fatty liver and is of great significance in reducing blood lipids in animals. It is a clinical treatment for non-alcoholic fatty liver and other fats. The metabolic disorders related diseases provide a theoretical basis. This project won the first prize of the 2017 National Undergraduate Life Science Innovation and Entrepreneurship Competition. The team members also published this SCI search paper in the New York Academy of Sciences Yearbook to remove iron-sensitive repressive ob/ob mice's non-alcoholic liver fat. Denaturation and its molecular mechanisms.

Iron is a basic element required for cell growth, but excessive iron can cause damage to the body. Iron ion chelating agents can significantly affect the level of iron inside and outside the cell, and are used clinically to treat various diseases caused by iron overload in human body. .

Nonalcoholic fatty liver disease is a clinical pathological syndrome characterized by no history of excessive drinking, characterized by hepatic steatosis and lipid storage. Epidemiological surveys show that in China, about 30% of adults have this disease. Studies have shown that serum iron levels in patients with nonalcoholic fatty liver disease increase, liver iron staining positive reaction is enhanced, and this disease is closely related to increased mortality associated with cardiovascular disease, liver and other tumors.

The classic iron chelator DFO is mainly used for the treatment of iron-loaded diseases such as thalassemia major, and has been clinically safe for decades. Xue Yu, a member of the undergraduate innovation team of the School of Life Science and Health, Northeastern University, under the guidance of the instructor Guo Wei, applied DFO to the ob/ob diabetes (obesity, overeating) model mice for intraperitoneal injection for the first time. It is confirmed that iron chelator has a clear alleviation effect on nonalcoholic fatty liver degeneration, and reducing specific tissue iron ions is an effective method for improving hepatic steatosis in ob/ob obese mice.

According to Xue Yu, the experimental data prove that iron chelator can reduce the iron deposition of liver tissue, reduce the level of oxidative stress, simultaneously up-regulate the expression level of lipid metabolism-related proteins, maintain the liver lipid metabolism and the homeostasis of iron metabolism, and then improve the liver. Steatosis. At the same time, iron chelator can inhibit the production of inflammatory cytokines and apoptosis of hepatocytes, up-regulate the expression level of hypoxia-inducible factor-1 and its related adaptive protein, and increase the ability of liver tissue to adapt and regenerate. In order to provide a new perspective for the clinical treatment of type II diabetes and non-alcoholic fatty liver disease with iron chelating agent "new drug".

It is reported that the reduction of dietary iron content, bloodletting therapy and other iron therapy are very effective in the treatment of nonalcoholic fatty liver, but these two methods are difficult to accurately control, making it difficult for ordinary patients to accept, so large-scale promotion is limited. Because the iron chelator DFO has been clinically safe for decades, it has a significant role in reducing animal blood lipids and hepatic steatosis. It can be promoted as a relatively safe and efficient drug, and has the potential to be rapidly transferred to clinical applications.
